    Abstract: This Test Guideline describes a method to evaluate photo-cytotoxicity by the relative reduction in viability of cells exposed to the chemical in the presence versus absence of light. Balb/c 3T3 cells are maintained in culture for 24 h for formation of monolayers. Two 96-well plates are pre-incubated with eight different concentrations of the test substance for 1 h. Thereafter one of the two plates is exposed to the highest non-cytotoxic irradiation dose whereas the other plate is kept in the dark. Cytotoxicity in this test is expressed as a concentration-dependent reduction of the uptake of the Vital dye Neutral Red (NR) when measured 24 hours after treatment with the test chemical and irradiation. NR penetrates cell membranes by non-diffusion, accumulating in lysosomes. Alterations of the cell surface of the sensitive lysosomal membrane lead to lysosomal fragility and other changes that gradually become irreversible. Such changes result in a decreased uptake and binding of NR. It is thus possible to distinguish between viable, damaged or dead cells. To predict the phototoxic potential, the concentration responses obtained in the presence and in the absence of irradiation are compared, usually at the IC50 level, i.e., the concentration reducing cell viability to 50 % compared to the untreated controls. The study report should include the development time and the total number of fully emerged midges (sex and number are recorded daily), the observation of any abnormal behaviour the number of visible pupae that have failed to emerge and any egg masses deposition. The data are analysed either by using a regression model in order to estimate the concentration that would cause x % reduction in emergence or larval survival or growth, or by using statistical hypothesis testing to determine a NOEC/LOEC.

Nevertheless, Spain faces important challenges related to high energy intensity, high water use intensity, and increasing CO2 emissions and municipal waste. This report makes 46 recommendations to help strengthen Spain’s environmental performance in the context of sustainable development.

The study report should include the adult morphological changes, the number of offspring. The reproductive output of the animals exposed to the test substance is compared to that of the control(s) in order to determine the no observed effect concentration (NOEC) and/or ECx by using a regression model to estimate the concentration that would cause a x % reduction in reproductive output. The test concentrations should bracket the ECx so that the ECx then comes from interpolation rather than extrapolation.

    Abstract: The method described in this Test Guideline is based on soil column chromatography in disturbed soil. Two types of experiments are performed to determine (i) the leaching potential of the test substance, and (ii) the leaching potential of transformation products in soils under controlled laboratory conditions. At least duplicate leaching columns are packed with untreated, air-dried and sieved soil (〈 2 mm) up to a height of approximately 30 cm. Afterwards they are saturated and equilibrated with an “artificial rain” solution and allowed to drain. Then the surface of each soil column is treated with the test substance (non-volatile in water and soil) and/or with aged residues of the test substance. Artificial rain is applied to the soil columns and the leachate is collected. After the leaching process the soil is removed from the columns and is sectioned into an appropriate number of segments depending on the information required from the study. A reference substance (atrazine or monuron) should be used in the leaching experiments. For each soil segment and leachate fraction, the amounts of test substance, transformation products, non-extractables and, if included, of the reference substance should be given in % of applied initial dose.

This Guideline is designed as a tiered approach; each tier is triggered by the results of the previous tier.Sterile aqueous buffer solutions of different pH values (pH 4, 7 and 9) are treated with the non-labelled or labelled test substance (only one concentration, which should not exceed 0.01 M or half of the saturation concentration). They are incubated in the dark under controlled laboratory conditions (at constant temperatures). After appropriate time intervals, buffer solutions are analysed for the test substance and for hydrolysis products. The preliminary test should be carried out for 5 days at 50 ± 0.5°C and pH 4.0, 7.0 and 9.0. The second tier consists of the hydrolysis of unstable substances, and the third tier is the identification of hydrolysis products. The higher Tier tests should be conducted until 90 % hydrolysis of the test substance is observed or for 30 days whichever comes first.

    Abstract: The purpose of this test is to measure the time course of biodegradation of a test substance at low concentration in aerobic natural water and to quantify the observations in the form of kinetic rate expressions. The test is performed in batch by “pelagic test” or “suspended sediment test” to simulate a water body with suspended solids or re-suspended sediment. The test flasks are incubated in darkness at an environmental temperature under aerobic conditions and agitation during 60 days normally. At least two different concentrations of test substance (non-volatile or slightly volatile organic) should be used. The maximum test concentration should be less than 100µg/L (biodegradation following first order kinetics) and the lowest test concentration preferably in the range of ‹1-10µg/L. Two subsamples should be withdrawn from each test flask at each sampling time. Degradation is followed at appropriate time intervals, by measuring either the residual 14C or the residual concentration of test substance. The total mineralisation and the primary biodegradation are determined by a different 14C labeling part of the molecule. Periodic measurements of pH and oxygen concentration in the test system must be conducted.

The chemicals are retained in the column in proportion to their hydrocarbon-water partition coefficient, with hydrophilic chemicals eluted first and lipophilic chemicals last. The HPLC method covers log Pow in the range of 0 to 6, but it can be expanded to cover the log Pow range between 6 and 10 in exceptional cases. The HPLC operation mode is isocratic. The test substance is injected in the smallest detectable quantities in the column. The retention time is determined in duplicate. The partition coefficient of the test substance is obtained by interpolation of the calculated capacity factor on the calibration graph. For very low and very high partition coefficients extrapolation is necessary.

    Abstract: This Test method has been designed to provide information on absorption of a test substance, (ideally radiolabelled), applied to the surface of a skin sample separating the two chambers (a donor chamber and a receptor chamber) of a diffusion cell. Static and flow-through diffusion cells are both acceptable. Skin from human or animal sources can be used. Although viable skin is preferred, non-viable skin can also be used. The skin has been shown to have the capability to metabolise some chemicals during percutaneous absorption. In this case, metabolites of the test chemical may be analysed by appropriate methods. Normally more than one concentration of the test substance is used in typical formulations, spanning the realistic range of potential human exposures. The application should mimic human exposure, normally 1-5 mg/cm2 of skin for a solid and up to 10 µl/cm2 for liquids. The temperature must be constant because it affects the passive diffusion of chemicals. The absorption of a test substance during a given time period (normally 24h) is measured by analysis of the receptor fluid and the distribution of the test substance chemical in the test system and the absorption profile with time should be presented.

    Abstract: The test described in this Test Guideline allows the identification of corrosive chemical substances and mixtures and it enables the identification of non-corrosive substances and mixtures when supported by a weight of evidence determination using other existing information. The test protocol may also provide an indication of the distinction between severe and less severe skin corrosives. This Test Guideline does not require the use of live animals or animal tissue for the assessment of skin corrosivity. The test material (solid or liquid) is applied uniformly and topically to a three-dimensional human skin model, comprising at least a reconstructed epidermis with a functional stratum corneum. Two tissue replicates are used for each treatment (exposure time), and for controls. Corrosive materials are identified by their ability to produce a decrease in cell viability below defined threshold levels at specified exposure periods. The principle of the human skin model assay is based on the hypothesis that corrosive chemicals are able to penetrate the stratum corneum by diffusion or erosion, and are cytotoxic to the underlying cell layers.

Immobilisation is recorded at 24 hours and 48 hours and compared with control values. The results are analysed in order to calculate the EC50 at 48h. Determination of the EC50 at 24h is optional. At least 2 ml of test solution should be provided for each animal (i.e. a volume of 10 ml for five daphnids per test vessel). The limit test corresponds to one dose level of 100 mg/L. The study report should include the observation for immobilized daphnids at 24 and 48 hours after the beginning of the test and the measures of dissolved oxygen, pH, concentration of the test substance, at the beginning and end of the test.

    Abstract: The Globally Harmonised System for the Classification and Labelling of Chemicals defines skin corrosion as the production of irreversible tissue damage in the skin following the application of a test material. The test material (150 µL for liquids or solid with 150 µL of deionised water added on the top) is applied for up to 24 hours to the epidermal surfaces of skin discs (three skin discs are used for each test and control substance) in a two-compartment test system in which the skin discs function as the separation between the compartments. The skin discs are taken from humanely killed rats aged 28-30 days. Corrosive materials are identified by their ability to produce a loss of normal stratum corneum integrity and barrier function, which is measured as a reduction in the TER below a threshold level (5kΩ for rat). A dye-binding step incorporated into the test procedure permits to determine if the increase in ionic permeability is due to physical destruction of the stratum corneum.

    Abstract: The in vivo percutaneous absorption study set out in this Test Guideline provides the linkage necessary to extrapolate from oral studies when making safety assessments following dermal exposure. The in vivo method, described in this guideline, allows the determination of the penetration of the test substance through the skin into the systemic compartment. The test substance, preferably radiolabelled, is applied, for a fixed period of time, to the clipped skin of animals at one or more appropriate dose levels in the form of a representative in-use preparation. The rat is the most commonly used species. At least four animals of one sex should be used for each test preparation and each scheduled termination time. A known amount of the test preparation is evenly applied to the site. This amount should normally mimic potential human exposure, typically 1-5 mg/cm² for a solid or up to 10 µl/cm² for liquids. A relevant exposure period (typically 6 or 24 hours) should be used, based on the expected human exposure duration. The animals should be observed for signs of toxicity/abnormal reactions at intervals for the entire duration of the study. This study includes: daily measurements (excreta), regular detailed observations, as well as sacrifice at the scheduled time and blood collected for analysis.

    Abstract: This method provides information on health hazard likely to arise from exposure to test substance (liquids, solids and aerosols) by application on the eye. This Test Guideline is intended preferably for use with albino rabbit. The test substance is applied in a single dose in the conjunctival sac of one eye of each animal. The other eye, which remains untreated, serves as a control. The initial test uses an animal; the dose level depends on the test substance nature. A confirmatory test should be made if a corrosive effect is not observed in the initial test, the irritant or negative response should be confirmed using up to two additional animals. It is recommended that it be conducted in a sequential manner in one animal at a time, rather than exposing the two additional animals simultaneously. The duration of the observation period should be sufficient to evaluate fully the magnitude and reversibility of the effects observed. The eyes should be examined at 1, 24, 48, and 72 hours after test substance application. The ocular irritation scores should be evaluated in conjunction with the nature and severity of lesions, and their reversibility or lack of reversibility. The individual scores do not represent an absolute standard for the irritant properties of a material, as other effects of the test material are also evaluated.

    Abstract: This Test Guideline describes a laboratory test method to assess aerobic and anaerobic transformation of organic chemicals in aquatic sediment systems. The method permits the measurement of (i) the transformation rate of the test substance in a water-sediment system and in the sediment (ii) the mineralisation rate of the test substance and/or its transformation products, (iii) the distribution of the test substance and its transformation products between the two phases during a period of incubation in the dark, at constant temperature, and (iv) the identification and quantification of transformation products in water and sediment phases including mass balance. At least two sediments different with respect to organic carbon content and texture are used. Ideally the test substance (one concentration) should be applied as an aqueous solution into the water phase. The duration of the experiment should normally not exceed 100 days, and should continue until the degradation pathway and water/sediment distribution pattern are established or when 90 % of the test substance has been removed by transformation and/or volatilisation. The number of sampling times should be at least six. The study includes: concentration in the water and sediment of the test substance and the transformation products at every sampling time; results from gases/volatiles trapping systems at each sampling time; mineralisation rates; and non-extractable residues in sediment at each sampling point. Half-lives, DT50, DT75 and DT90 values are determined where the data warrant.

The experiments are performed to determine the rate of transformation of the test substance, and the nature and rates of formation and decline of transformation products, to which plants and soil organisms may be exposed. About 50 to 200 g soil samples (a sandy loam or silty loam or loam or loamy sand) are treated with the test substance and incubated in the dark, in biometer-type flasks or in flow-through systems under controlled laboratory conditions. The treatment rate should correspond to the highest application rate of a crop protection product recommended in the use instructions. Also untreated soil samples are incubated under test conditions. These samples are used for biomass measurements during and at the end of the studies. The rate and pathway studies should normally not exceed 120 days. Duplicate incubation flasks are removed at appropriate time intervals and the soil samples extracted with appropriate solvents, of different polarity, and analysed for the test substance and/or transformation products. Volatile products are also collected for analysis using appropriate adsorption devices. Using 14C-labelled material, the various mineralisation rates of the test substance can be measured by trapping evolved 14CO2 and a mass balance, including the formation of soil bound residues, can be established.

This Test Guideline recommends sequential testing strategies, which include the performance of validated and accepted in vitro or ex vivo tests for corrosion/irritation. The albino rabbit is the preferable laboratory animal. The substance to be tested is applied in a single dose to a small area of skin (approximately 6cm²) of an experimental animal; untreated skin areas of the test animal serve as the control. The exposure period is 4 hours. Residual test substance should then be removed. The dose is 0.5ml (liquid) or 0.5g (solid) applied to the test site. The method consists of two tests: the initial test and the confirmatory test (used only if a corrosive effect is not observed in the initial test). All animals should be examined for signs of erythema and oedema during 14 days. The dermal irritation scores should be evaluated in conjunction with the nature and severity of lesions, and their reversibility or lack of reversibility. When responses persist to the end of the 14-day observation period, the test substance should be considered an irritant.

    Abstract: A principle of the method is that in the main study only moderately toxic doses are used, and the administration of doses that are expected to be lethal should be avoided. This Guideline is intended primarily for use with rat. Groups of animals of a single sex (normally females) are dosed in a stepwise procedure using the fixed doses of 5, 50, 300 and 2000 mg/kg (exceptionally 5000 mg/kg). The initial dose level is selected on the basis of a sighting study as the dose expected to produce some signs of toxicity without causing severe toxic effects or mortality. Further groups of animals may be dosed at higher or lower fixed doses, depending on the presence or absence of signs of toxicity or mortality. This procedure continues until the dose causing evident toxicity or death is identified, or when no effects are seen at the highest dose or when deaths occur at the lowest dose. The test substance is administered in a single dose by gavage using a stomach tube or a suitable intubation canula. Animals should be fasted prior to dosing. A total of five animals of one sex will normally be used for each dose level investigated. The results of this study include: measurements (weighing at least weekly) and daily detailed observations, as well as gross necropsy. The method provides information on the hazardous properties and allows the substance to be classified for acute toxicity according to the Globally Harmonised System of classification and labelling of chemicals.

    Abstract: This Test Guideline describes two simulations tests used in the aerobic sewage treatment. Activated Sludge Units are designed to determine the elimination and the primary and/or ultimate biodegradation of water-soluble organic compounds by aerobic micro-organisms in a continuously operated test system simulating the activated sludge process. Two continuously operated test units are run in parallel under identical conditions. Normally the mean hydraulic retention time is 6 h and the mean sludge age is 6 to 10 days. Sludge is wasted by one of two methods, the test substance is normally added at a concentration of between 10 mg/l dissolved organic carbon (DOC) and 20 mg/l DOC, to the influent of only one of the units. The second unit is used as a control. The DOC, preferably, or chemical oxygen demand (COD) is determined, together with the concentration of the test substance by specific analysis, in the effluent from the unit receiving the test substance. In Biofilms, synthetic or domestic sewage, and the test substance, in admixture or alone, are applied to the internal surface of a slowly rotating inclined tube. A layer of microorganisms is built up on the internal surface. Effluent from the tube is collected and either settled and/or filtered before analysis for DOC and/or the test substance by a specific method. Control units are operated in parallel under the same conditions. The difference between the concentrations of DOC/COD in the effluent from the test and control units is assumed to be due to the test substance. This difference is compared with the concentration of the added test substance to calculate the elimination of the test substance. Biodegradation may normally be distinguished from bio-adsorption by careful examination of the elimination-time curve.

    Abstract: This Test Guideline for two-generation reproduction testing is designed to provide general information concerning the effects of a test substance on the integrity and performance of the male and female reproductive systems, and on the growth and development of the offspring. The test substance is administered daily in graduated doses to several groups of males and females. Males and females of the Parent generation (5-9 weeks old) should be dosed during growth, during their mating, during the resulting pregnancies, and through the weaning of their first generation offspring. The administration of the substance is continued to first generation offspring during their growth into adulthood, mating and production of a second generation (until the weaning). The rat is the preferred species for testing. Each test and control group should contain a sufficient number of animals to yield preferably not less than 20 pregnant females at or near parturition. At least three dose levels and a concurrent control shall be used. It is recommended that the test substance be administered orally (by diet, drinking water or gavage). A limit test may be performed if no effects would be expected at a dose of 1000 mg/kg bw/d. The results of this study include: measurements (weighing, sperm parameters, oestrus cycle parameters and offspring parameters), clinical daily observations, as well as gross necropsy and histopathology. The findings of this two-generation reproduction toxicity study should be evaluated in terms of the observed effects including necropsy and microscopic findings. A properly conducted reproductive toxicity test should provide a satisfactory estimation of a no-effect level and an understanding of adverse effects on reproduction, parturition, lactation, postnatal development including growth and sexual development.

Particular focus is placed on how the use of the price system, reform of governments' decision-making processes, technology policies, and international trade and investment may each contribute to removing incentives to unsustainable resource depletion and environmental degradation. Recommendations in each of these areas are presented in this Report, and then applied to two areas -- climate change and natural resource management -- where the risks of non-sustainability appear to be highest.

    Abstract: This Test Guideline for developmental toxicity testing is designed to provide general information concerning the effects of prenatal exposure on the pregnant test animal and on the developing organism. The test substance is normally administered to pregnant animals at least from implantation to one day prior to the day of scheduled kill, which should be as close as possible to the normal day of delivery. This Test Guideline is intended for use with rodent (rat preferably) and non-rodent (rabbit preferably). Each test and control group should contain a sufficient number of females to result in approximately 20 female animals with implantation sites at necropsy. Three concentrations, at least, should be used. The test substance or vehicle is usually administered orally by intubation. A limit test may be performed if no effects would be expected at a dose of 1000 mg/kg bw/d. The results of this study include measurements (weighing) and clinical daily observations, each day preferably at the same time. Shortly before caesarean section, the females are killed (one day prior to the expected day of delivery), the uterine contents are examined, and the foetuses are evaluated for soft tissue and skeletal changes. A number of endocrine-related measurements in the dams and in the fetuses have been added in 2018. In any study which demonstrates an absence of toxic effects, further investigation to establish absorption and bioavailability of the test substance should be considered.

    Abstract: This Test Guideline describes an experimental method using HPLC for the estimation of the adsorption coefficient Koc in soil and in sewage sludge. The HPLC dead time t0 may be determined by two methods: the determination by means of a homologous series and the determination by inert substances which are not retained by the column. If the actual results show that the log Koc of the test substance is outside the calibration range the test should be repeated using different, more appropriate reference substances.

    Abstract: On the surface there is agreement, sustainable development refers to a broad set of issues, going beyond the relationship between the economy and the environment to encompass human and social concerns. Scratch the surface and you open a Pandora's box of differing notions of sustainability and means of achieving it. How can progress towards sustainable development be measured then? The major difficulty in developing indicators to track progress towards sustainable development is not the lack of data but rather the lack of frameworks to organise and synthesize existing information.     Abstract: This Test Guideline describes a laboratory test method designed to investigate long term potential effects of a single exposure of agrochemicals/non agrochemicals on carbon transformation activity of soil microorganisms. A minimum of two test concentrations are recommended for agrochemicals (five for non agrochemicals). Sieved soil is divided into portions of equal weight (three for agrochemicals, six for non agrochemicals) including portions mixed with the carrier containing the product, and the control. A minimum of three replicates for both treated and untreated soils is recommended. After 0, 7, 14 and 28 days incubation, samples of treated and control soils are mixed with glucose, and glucose-induced respiration rates are measured for 12 consecutive hours. Respiration rates are expressed as carbon dioxide released or oxygen consumed. The mean respiration rate in the treated soil samples is compared with that in control and the percent deviation of the treated from the control is calculated. All tests run for at least 28 days. If, on the 28th day, differences between treated and untreated soils are equal to or greater than 25 % measurements are continued in 14 day intervals for a maximum of 100 days. Results are analysed using a regression model, and the ECx values are calculated. When the difference in respiration rates between the lower treatment and control is equal to or less than 25 % at any sampling time after day 28, agrochemicals can be evaluated as having no long-term influence on carbon transformation in soils. The EC50, EC25 and/or EC10 values are used for non agrochemicals.

    Abstract: This Test Guideline describes a method to analyse the solution/extraction behaviour of polymers in water, which uses the flask method with modifications. Three samples of 10 g of particles of the test substance are weighed into each of three vessels fitted with glass stoppers. 1000 ml of water is added to each vessel. The vessels are tightly stoppered and then agitated at 20 °C normally during a period of 24 hours. Then, the content of each vessel is centrifuged or filtered and the concentration of polymer in the clear aqueous phase is determined by a suitable analytical method. For a direct analysis of the aquous phase one sample size is sufficent. Analysis of the aqueous phase for the total polymeric species is possible either by a sufficiently sensitive method or by vacuum evaporation to dryness of the aqueous extract and spectroscopic or AAS/ICP analysis of the residue. When determination of the solution/extraction behaviour of the polymer consists in determining only the total organic carbon content (TOC) of the aqueous extract, an additional test should be conducted, with two different sample sizes in triplicate, using ten times smaller polymer samples and the same amounts of water as those used in the first test. A gravimetric determination is performed on the undissolved/not-extracted part of the sample.

    Abstract: This Test Guideline is aimed at estimating the adsorption/desorption behaviour of a chemical on different soil types. The goal is to obtain a sorption value which can be used to predict partitioning under a variety of environmental conditions; to this end, equilibrium adsorption coefficients for a chemical on various soils are determined as a function of soil characteristics (organic carbon, clay content, soil texture, and pH). The test comprises three tiers. The tier 1 is the preliminary study, the tier 2 is the screening test (in 5 soils) and the tier 3 is the determination of Freundlich adsorption isotherms or the study of desorption by means of desorption kinetics/Freundlich desorption isotherms, as appropriate. Two methods are possible for analyse: the indirect method and the direct method. The indirect method consists of the adjunction of the test substance to soil samples, the agitation of the mixture for an appropriate time, the analysis of the aqueous phase after centrifugation and the filtration of the soil suspension. The amount of test substance adsorbed on the soil sample is calculated as the difference between the amount of test substance initially present in solution and the amount remaining at the end of the experiment. The direct method is recommended when the difference in the solution concentration of the substance cannot be accurately determined.
