    Kurzfassung: This method provides information on health hazard likely to arise from short-term exposure to test substance. The determination of sub-chronic oral toxicity using repeated doses may be carried out after initial information on toxicity has been obtained from acute or repeated dose 28-day toxicity tests. The method is based on the repeated oral administration of the substance of interest over a prolonged period (one dose level daily during 90 days). This Test Guideline is intended primarily for use with non-rodents species. The commonly used non-rodent species is the dog (the beagle is frequently used). At least 8 animals (4 female and 4 male) should be used for each test group. Three concentrations, at least, should be used. The test compound is administered in the diet or in the drinking water, by gavage or in capsules. A limit test may be performed if no effects would be expected at a dose of 1000 mg/kg bw/d. The results of this study include: measurements (weighing at least once a week, food/water consumption) and daily (preferably at the same time) and detailed observations (ophtalmological examination, haematology, clinical biochemistry and urinalysis), as well as gross necropsy and histopathology. A properly conducted 90-day subchronic test should provide a satisfactory estimation of a no-effect level.

    Kurzfassung: This Test Guideline is a laboratory test method, designed to assess the acute contact toxicity of pesticides and other chemicals to adult worker honeybees. Anaesthetized adult worker honeybees are exposed to five doses in a geometric series of the test substance dissolved in appropriate carrier (in total a volume of 1 ml), by direct application to the thorax (droplets). A minimum of three replicate test groups, each of ten bees, should be dosed with each test concentration. A toxic standard (usually dimethoate) should be included in the test series. The limit test corresponds to one dose level of 100 ìg active ingredient/bee. The test duration is 48h. Mortality is recorded daily during at least 48 hours and compared with control values. If the mortality rate is increasing between 24 and 48h whilst control mortality remains at an accepted level, it is appropriate to extend the duration of the test to a maximum of 96h. The results are analysed in order to calculate the LD50 at 24h and 48h and, in case the study is prolonged, at 72h and 96h.

    Kurzfassung: Cet essai de toxicité à court terme chez le poisson aux stades de l'embryon et de l'alevin consiste à exposer les stades compris entre l'oeuf qui vient d'être fécondé et la fin du stade de l'alevin. Les stades de l'embryon et de l’alevin des poissons sont exposés à cinq concentrations de la substance d'essai dissoute dans l'eau. Selon la nature de la substance d'essai, il est possible d’utiliser un essai semi-statique ou dynamique. L'essai débute quand on place au moins trente oeufs fécondés divisés en nombre égal entre au moins trois compartiments d’essai identiques pour chaque concentration. Il se termine juste avant que le sac vitellin d’une quelconque larve d’un quelconque compartiment ait été complètement absorbé ou avant que les poissons témoins ne meurent d’inanition. Les effets létaux et sublétaux sont évalués et comparés aux valeurs contrôles pour déterminer la concentration minimale avec effet observé et la concentration sans effet observé. Alternativement, ils peuvent être analysés en utilisant un modèle de régression afin d'estimer la concentration qui provoquerait un certain pourcentage d'effet. Le rapport d'étude inclut le comptage quotidien de la progéniture, l'enregistrement quotidien de la mortalité parentale, la mesure hebdomadaire de la concentration en oxygène, de la température, des valeurs du pH, et la détermination des concentrations de la substance d'essai. Elle devrait également inclure les observations d’apparence anormale, de comportement anormal, de l’éclosion et de la survie.

    Kurzfassung: In this Short-term Toxicity Test on Fish Embryo and Sac-Fry Stages, the life stages from the newly fertilized egg to the end of the sac-fry stage are exposed. The embryo and sac-fry stages of fish are exposed to five concentrations of the test substance dissolved in water. A choice, depending on the nature of the test substance, is possible between a semi-static and a flow-through procedure. The test starts with placing at least 30 fertilised eggs divided equally between at least three replicate test chambers per concentration, and is terminated just before the yolk-sac of any larvae in any of the test chambers has been completely absorbed or before mortalities by starvation start in controls. Lethal and sub-lethal effects are assessed and compared with control values to determine the lowest observed effect concentration and hence the no observed effect concentration. Alternatively, they may be analysed using a regression model in order to estimate the concentration that would cause a given percentage effect. The study report should include: the daily counting of the offspring, the daily recording of the parent mortality, the weekly measurement of oxygen concentration, temperature, and pH values; and the determination of the concentrations of test substance. Le composant testé est administré par gavage, dans des capsules ou via la nourriture ou la boisson. Un essai limite peut être fait quand on n’attend pas d’effet pour une dose de 1000 mg/kg poids corporel/j. Les résultats de l’étude incluent des mesures (le poids au moins une fois par semaine, la consommation de nourriture et d’eau), des observations quotidiennes détaillées (examen ophtalmologique, hématologie, biochimie clinique et analyse d’urine), chaque jour de préférence au même moment, de même qu’une autopsie générale et de l’histopathologie. Un essai sub-chronique de 90 jours correctement effectué devrait fournir une évaluation satisfaisante d'un niveau sans effet.

    Kurzfassung: This method provides information on health hazard likely to arise from exposure to test substance via oral administration. The determination of sub-chronic oral toxicity using repeated doses may be carried out after initial information on toxicity has been obtained from acute or repeated dose 28-day toxicity tests. The method is based on the repeated oral administration of the substance of interest over a prolonged period (one dose level daily during 90 days). This Test Guideline is intended primarily for use with rodents (rat preferably). At least 20 animals (10 female and 10 male) should be used for each test group. Three concentrations, at least, should be used. The test compound is administered by gavage or via the diet or drinking water. A limit test may be performed if no effects would be expected at a dose of 1000 mg/kg bw/d. The results of this study include: measurements (weighing at least once a week, food and water consumption) and daily and detailed observations (ophtalmological examination, haematology, clinical biochemistry and urinalysis), each day preferably at the same time; as well as gross necropsy and histopathology. A number of endocrine-related measurements, particularly relevant to thyoid function have been added in 2018. A properly conducted 90-day subchronic test should provide a satisfactory estimation of a no-effect level.

    Kurzfassung: This Test Guideline is a laboratory test method, designed to assess the oral acute toxicity of pesticides and other chemicals, to adult worker honeybees. Adult worker honeybees are exposed to five doses in a geometric series of the test substance dispersed in sucrose solution. A minimum of three replicate test groups, each of ten bees, should be dosed with each test concentration. A toxic standard (usually dimethoate) should be included in the test series. The bees are then fed the same diet, free of the test substance. The limit test corresponds to one dose level of 100 ìg active ingredient/bee. Mortality is recorded daily during at least 48 hours and compared with control values. If the mortality rate is increasing between 24 and 48h whilst control mortality remains at an accepted level it is appropriate to extend the duration of the test to a maximum of 96h. The study report should include the amount of diet consumed per group and the observation of all abnormal behavioural. The results are analysed in order to calculate the LD50 at 24h and 48h and, in case the study is prolonged, at 72h and 96h.

    Kurzfassung: Le but de l'essai synthèse non programmée de d'ADN (UDS) sur les cellules de foie de mammifères in vivo est d'identifier les substances qui induisent la réparation de l'ADN après excision et élimination d'un segment d'ADN contenant la région endommagée par l'agent physique ou chimique (solides ou liquides). L'essai est habituellement basé sur l'incorporation de thymidine tritiée, 3H-TdR, (pendant 3-8 heures) dans l'ADN des hépatocytes qui ont peu fréquemment des cellules en phase S du cycle de cellules. L’incorporation de 3H-TdR est habituellement déterminée par autoradiographie. Les rats sont généralement utilisés, et au moins trois animaux analysables par groupe sont utilisés. Normalement, au moins deux niveaux de dose sont employés. Un essai limite peut être réalisé si aucun effet n’est prévu à une dose de 2000 mg/kg pc/j. La substance d’essai est généralement administrée comme traitement unique par gavage en utilisant une sonde gastrique ou une canule d'intubation appropriée. Des hépatocytes sont prélevés sur des animaux traités 12-16 heures après l’administration du produit. Après l'autoradiographie, normalement 100 cellules sont comptées par animal sur au moins deux lames. Un résultat positif de l'essai d'UDS avec les cellules de foie mammifères indique in vivo qu'une substance induit des dommages d'ADN en cellules de foie mammifères in vivo qui peuvent être réparées par la synthèse imprévue d'ADN in vitro. Un résultat négatif indique que, dans les conditions d'essai, la substance d'essai n'induit pas des dommages d'ADN qui sont discernables par cet essai.

When the study is conducted as a separate study, at least 20 animals (10 females and 10 males) should be used in each dose. At least three dose groups and a control group should generally be used. Dose levels should be selected by taking into account any previously observed toxicity and kinetic data available for the test compound or related materials. The dosing regimen may be 28 days, subchronic (90 days) or chronic (1 year or longer). The procedures set out in this Test Guideline may also be used for an acute neurotoxicity study. The limit test corresponds to one dose level of at least 1000 mg/kg body weight. The results of this study include measurements (weighing, food /water consumption), functional tests, and, at least, daily detailed observations (Ophthalmology, haematology, clinical biochemistry and histopathology). At least five males and five females, selected from test group, should be perfused in situ and used for detailed neurohistopathology at the end of the study. The findings of the study should be evaluated in terms of the incidence, severity and correlation of neurobehavioural and neuropathological effects (neurochemical or electrophysiological effects as well if supplementary examinations are included) and any other adverse effects observed.

Test substances are preferably administered once or twice but they may also be administered as a split dose to facilitate administering a large volume of material. Prior to sacrifice, animals are treated with a metaphase-arresting agent. Chromosome preparations are then made from germ cells and stained, and metaphase cells are analyzed for chromosome aberrations. A limit test may be performed if no effects would be expected at a dose of 2000 mg/kg bw/d. Positive results from the in vivo spermatogonial chromosome aberration test indicate that a substance induces chromosome aberrations in the germ cells of the species tested.

Les cultures traitées sont maintenues dans le milieu de croissance pendant une période suffisante, caractéristique de chaque locus et de chaque type cellulaire, afin de permettre l'expression phénotypique presque optimale des mutations induites. La fréquence de mutant est déterminée par l’ensemencement d’un nombre connu de cellules dans le milieu contenant l'agent sélectif pour détecter des cellules mutantes, et dans le milieu sans agent sélectif pour déterminer l'efficacité de clonage (viabilité). La concentration d'essai maximum recommandée pour les substances non-cytotoxiques solubles est de 5 mg/boîte ou de 5 ml/boîte. Il y a deux méthodes : la méthode par étalement et la méthode de pré-incubation. Pour les deux techniques, après deux ou trois jours d'incubation à 37°C, les colonies révertantes sont comptées et comparées au nombre de colonies révertantes spontanées des boîtes de contrôle.

There are two methods: the plate incorporation method and the preincubation method. For both techniques, after two or three days of incubation at 37°C, revertant colonies are counted and compared to the number of spontaneous revertant colonies on solvent control plates.

    Kurzfassung: The purpose of the unscheduled DNA synthesis (UDS) test with mammalian liver cells in vivo is to identify substances that induce DNA repair after excision and removal of a stretch of DNA containing a region of damage induced by chemical substances (solid or liquid) or physical agents in the liver. The test is usually based on the incorporation of tritium-labelled thymidine, 3H-TdR, (during 3-8 hours) into the DNA of liver cells which have a low frequency of cells in the S-phase of the cell cycle. The uptake of 3H-TdR is usually determined by autoradiography. Rats are commonly used, and the number of animals should be at least three analysable animals per group. Normally, at least two dose levels are used. A limit test may be performed if no effects would be expected at a dose of 2000 mg/kg bw/d. Test substances are generally administered as a single treatment by gavage using a stomach tube or a suitable intubation cannula. Liver cells are prepared from treated animals 12-16 hours after dosing of animal. After autoradiography, normally 100 cells are scored from each animal from at least two slides. A positive result from the UDS test with mammalian liver cells in vivo indicates that a substance induces DNA damage in mammalian liver cells in vivo that can be repaired by unscheduled DNA synthesis in vitro. A negative result indicates that, under the test conditions, the test substance does not induce DNA damage that is detectable by this test.

In the cell lines the most commonly-used genetic endpoints measure mutation at thymidine kinase (TK) and hypoxanthine-guanine phosphoribosyl transferase (HPRT), and a transgene of xanthineguanine phosphoribosyl transferase (XPRT). The TK, HPRT and XPRT mutation tests detect different spectra of genetic events. Cells in suspension or monolayer culture are exposed to, at least four analysable concentrations of the test substance, both with and without metabolic activation, for a suitable period of time. They are subcultured to determine cytotoxicity and to allow phenotypic expression prior to mutant selection. It is recommended to utilise at least 106cells. Cytotoxicity is usually determined by measuring the relative cloning efficiency (survival) or relative total growth of the cultures after the treatment period. The treated cultures are maintained in growth medium for a sufficient period of time, characteristic of each selected locus and cell type, to allow near-optimal phenotypic expression of induced mutations. Mutant frequency is determined by seeding known numbers of cells in medium containing the selective agent to detect mutant cells, and in medium without selective agent to determine the cloning efficiency (viability). After a suitable incubation time, colonies are counted.

Bone marrow and/or blood cells are collected, prepared and stained. Preparations are analyzed for the presence of micronuclei. Each treated and control group must include at least 5 analysable animals per sex. Administration of the treatments consists of a single dose of test substance or two daily doses (or more). The limit dose is 2000 mg/kg/body weight/day for treatment up to 14 days, and 1000 mg/kg/body weight/day for treatment longer than 14 days.

    Kurzfassung: The purpose of the in vitro chromosome aberration test is to identify agents that cause structural chromosome aberrations in cultured mammalian somatic cells. Structural aberrations may be of two types: chromosome or chromatid. The in vitro chromosome aberration test may employ cultures of established cell lines, cell strains or primary cell cultures. Cell cultures are exposed to the test substance (liquid or solid) both with and without metabolic activation during about 1.5 normal cell cycle lengths. At least three analysable concentrations of the test substance should be used. At each concentration duplicate cultures should normally be used. At predetermined intervals after exposure of cell cultures to the test substance, the cells are treated with a metaphase-arresting substance, harvested, stained. Metaphase cells are analysed microscopically for the presence of chromosome aberrations.

    Kurzfassung: Cet essai mesure des événements chromosomiques dans les cellules germinales de spermatogonies et devrait donc prévoir l'induction de mutations héréditaires dans les cellules germinales. Des hamsters chinois et des souris mâles sont généralement utilisés. Les animaux sont exposés à la substance d'essai (liquide ou solide) par une voie appropriée d'exposition, habituellement par gavage ou par injection intrapéritonéale. Puis, ils sont sacrifiés à des moments appropriés après traitement. Chacun des groupes traités et témoins incluent au moins cinq mâles analysables. Les substances d'essai sont de préférence administrées une ou deux fois mais elles peuvent également être administrées en prises fractionnées quand le volume à administrer est grand. Avant le sacrifice, les animaux sont traités avec un inhibiteur de fuseau. Des préparations chromosomiques sont alors faites à partir des cellules germinales puis colorées. Les aberrations chromosomiques des cellules en métaphase sont analysées. Un essai limite peut être réalisé si aucun effet n’est prévu à une dose de 2000 mg/kg pc/j. Des résultats positifs de l'essai d'aberration chromosomique sur des spermatogonies in vivo indiquent qu'une substance induit des aberrations chromosomiques dans les cellules germinales des espèces examinées.

Des niveaux de dose devraient être choisis en prenant en considération toute toxicité précédemment observée et toutes données cinétiques disponibles pour la substance d'essai. Le régime de dosage peut être de 28 jours, 90 jours (subchronique) ou 1 an ou plus (chronique). Les procédures visées dans cette Ligne directrice peuvent également être employées pour une étude aiguë de neurotoxicité. L'essai de limite correspond à un niveau de dose d’au moins 1000 mg/kg pc. Les résultats de cette étude incluent des mesures (pesée, consommation d’eau et de nourriture), des essais fonctionnels, et des observations, au moins quotidiennes, détaillées (ophtalmologie, hématologie, biochimie clinique et histopathologie).     Kurzfassung: This Test guideline describes the Gel Permeation Chromatography (GPC). This method permits to determine the molecular weight distribution and the average molecular weights (Mn, Mw). GPC is a special type of liquid chromatography in which the sample is separated according to the hydrodynamic volumes of the individual constituents.According to their size, the eluted molecules can or not penetrate in the porous material (typically an organic gel) of which the columns are filled. Thus, the smallest molecules are retained whereas largest elute more quickly. At exit of column, detectors (generally by differential refractometry) provide the refractive index or UV-absorption and yield a simple distribution curve. However, to attribute actual molecular weight values to the curve, it is necessary to calibrate the column by passing down polymers of known molecular weight and, ideally, of broadly similar structure, e.g. various polystyrene standards. For each sample analyzed, two independent experiments must be undertaken. They have to be analysed individually. Mn, Mw, Mw/Mn must be provided for every measurement.

    Kurzfassung: This Test Guideline describes a procedure for characterising the bioconcentration potential of substances in fish, under flow-through conditions (but semi-static regimes are permissible). The test consists of two phases: the exposure (uptake) and post-exposure (depuration) phases. During the uptake phase (28 days normally and 60 days maximum), separate groups of four fishes of one species are exposed to at least two concentrations of the test substance. They are then transferred to a medium free of the test substance for the depuration phase. A depuration phase is always necessary unless uptake of the substance during the uptake phase has been insignificant. In addition to the two test concentrations, a control group of fish is held without the test substance. The concentration of the test substance in/on the fish is followed through both phases of the test. During the test, dissolved oxygen, TOC, pH, total hardness and salinity, and temperature should be measured in vessels. The lipid content should be determined on the same biological material as is used to determine the concentration of the test substance, when feasible. Where possible the bioconcentration factor at apparent steady-state (BCF), expressed as a function of the total wet weight of the fish, and the kinetic bioconcentration factor (BCFK) are calculated. The bioconcentration should be expressed in relation to lipid content in addition to whole body weight.

Il est recommandé que la substance d’essai soit administrée oralement par gavage. Ceci devrait être fait avec une dose unique quotidienne en utilisant une sonde gastrique ou une canule d'intubation appropriée. Chaque groupe devrait initialement inclure au moins 10 animaux de chaque sexe. Généralement, au moins trois groupes d'essai et un groupe de contrôle devraient être employés. Des niveaux de dose devraient être choisis, en tenant compte de toutes données de toxicité existantes et de (toxico-) cinétique disponibles.     Kurzfassung: The test substance is administered in graduated doses to several groups of males and females. Males should be dosed for a minimum of four weeks; females should be dosed throughout the study (approximately 54 days). Normally, matings “one male to one female” should be used in this study. This Test Guideline is designed for use with the rat. It is recommended that the test substance be administered orally by gavage. This should be done in a single dose daily to the animals using a stomach tube or a suitable intubation cannula. Each group should be started with at least 10 animals of each sex. Generally, at least three test groups and a control group should be used. Dose levels should be selected taking into account any existing toxicity and (toxico-) kinetic data available. The limit test corresponds to one dose level of at least 1000 mg/kg body weight. The results of this study include measurements (weighing, food/water consumption) and daily detailed observations (including sensory reactivity to stimuli), preferably each day at the same time, as well as gross necropsy and histopathology. The findings of this toxicity study should be evaluated in terms of the observed effects, necropsy and microscopic findings. The evaluation will include the relationship between the dose of the test substance and the presence or absence of observations. Because of the short period of treatment of the male, the histopathology of the testis and epididymus must be considered along with the fertility data, when assessing male reproduction effects.
