The Special Issue is dedicated to Professor Gerda van Rosmalen who was a pioneer in this field.     Abstract: This reprint focuses on a very important topic in metrology, which is represent by measurement uncertainty. Any good metrologist or scientist in engineering knows that no measurement makes sense without an associated uncertainty value: without an uncertainty value, no decision can be taken; no comparisons can be made; no conformity can be assessed. Any decision, comparison or conformity assessment made without considering the measurement uncertainty affecting the measurement value is completely useless and meaningless. Stated that, it becomes very clear that uncertainty in measurement plays indeed a very important rule in our everyday life. This is the reason why there is a great need to have a fruitful academic and scientific discussion on this topic. We have been speaking about measurement uncertainty for less than 30 years, since the concept of “measurement uncertainty” has been introduced in 1995 by the “Guide to the expression of uncertainty in measurement” (GUM). Thirty years seems to be many, but still the concept of measurement uncertainty has not been spread worldwide and the GUM is a document that is not known everywhere. On the other hand, this document should be considered not only in academic scenario, but also in any technical and industrial scenario, where it is pivotal to know the meaning of measurement uncertainty, identify the uncertainty contributions and know how these contributions affect the final measurement result

    Abstract: The importance of studying pollution via potentially toxic elements (PTEs) is becoming a key challenge considering the increasing concern about the presence of PTEs in the environment and their potential impacts on ecosystem functioning and public health due to their persistence and biotoxicity. Regional geochemical research, together with geostatistical computations which are used to identify source patterns of different pollutants related to the underlying geological features and/or anthropogenic activities, is the best approach for characterizing this pollution issue. The collection of research in this Special Issue, “Potentially Toxic Element Pollution in Urban and Suburban Environments”, represents the need of the scientific community to characterize the behavior, transport, fate, and ecotoxicological state of PTEs in environmental matrices in both urban and suburban settings.     Abstract: Halophytes are a fascinating group of wild plants adapted to highly saline natural habitats, where most plant species and all our conventional crops would not survive. In fact, some halophytes can withstand even seawater salinity. In the current climate change scenario, increasing average temperatures and drought episodes contribute to the accelerated salinisation of irrigated cropland, especially in arid and semiarid regions, by the progressive accumulation in the soil of salts dissolved in irrigation water. This ‘secondary salinisation’ is one of the major causes of reducing crop yields worldwide. In this context, halophytes represent ideal experimental systems to investigate the mechanisms plants use to respond to high-salinity conditions. This knowledge will be essential for the genetic improvement of crop salt tolerance, which represents the most sensible strategy to address the abovementioned problem. Furthermore, halophytes could be the basis of a sustainable, ‘saline’ agriculture, after domestication and some breeding to improve agronomic characteristics. Then, they could be commercially cultivated for food, feed, fibre, or the production of biomolecules of industrial interest. Since they could be grown in saline land and irrigated with brackish water, they will not compete with our conventional crops for these limited resources, fertile land and good-quality water for irrigation. The articles included in this Special Issue address these different aspects of halophytes’ research, although most focus on basic studies on salt-tolerance mechanisms

    Abstract: Data have become an immensely valuable resource. They are the key driver that puts the smart into smart services. Over the last two decades, sophisticated OMICs technologies—genomics, transcriptomics, proteomics, peptidomics, metabolomics and lipidomics, also known as foodomics—have been powerful approaches that extended the scope of traditional methods and have established impressive possibilities of addressing meat quality issues. Foodomics were further used to elucidate the biological basis/mechanisms of phenotypic variation in the technological and sensory quality traits of meat from different species. Overall, these techniques aimed to comprehensively study the dynamic link(s) between the genome and the quality traits of the meat that we eat compared to traditional methods, hence improving both the accuracy and sensitivity thanks to the large quantities of data that can be generated. This Special Issue focused on the cutting-edge research applications of OMICs tools to characterize or manage the quality of muscle foods. Moreover, the coastal region is home to a significant number of distinct biological communities, including coral reefs, mangroves, salt meadows and wetlands, phanerogam meadows, and kelp forests, estuarine assemblages or coastal lagoons, forests, and grasslands. Remote sensing meets this challenge by offering a wide range of standard products on environmental coastal condition, thanks in particular to various state-of-the-art sensors. The development of innovative methods based on the integration of multi-source, multi-resolution, and multi-temporal images offers promising prospects for considering the different scales of ecosystems.     Abstract: Mangrove communities represent a coastal habitat located in the intertidal zone or brackish water of tropical and subtropical coastal areas, between 5°N and 5°S latitudes, spanning over 118 countries. The special ecological conditions of mangroves include high salinity, nutrient limitation, tidal gradients, high temperatures, excessively high light, and muddy anaerobic or sandy soil, which lead to various morphological and physiological adaptations of inhabiting species and act as an effective selector for the metabolic pathway via the generation of unique functional metabolites with highly unique chemical scaffolds and pharmaceutical application potential. In recent decades, numerous metabolites with uncommon structures and efficacious bioactivities have been discovered in mangrove-derived microorganisms, along with mangrove plants. For this reason, mangrove ecosystems have taken the limelight as an attractive biodiversity hotspot, attracting significant attention from organic chemists and pharmacologists. Additionally, many unique and novel chemical structures with a wide range of structural classes have suggested various biosynthetic origins containing novel functional genes and corresponding enzymes with unique catalytic functions. The scope of this reprint embraces numerical work on material responses to extreme conditions, such as high-speed impact or loading, neutron or ion irradiation, and high-pressure and/or high-temperature environments. Related simulation results based on the first principle method, molecular dynamics, and finite element methods are reported. Results of various topics have been reported by papers collected in this book, for example, including fitting a better empirical potential, diffusion of atoms on surface, corrosion, interaction between different defects, elastic constants calculations, effect of hot press forming on collision toughness and energy distribution, and influence of radiation defects on the thermo-mechanical properties of UO2. Furthermore, a review about the experiments and models for the effect of strain rate on NiTi shape memory alloys (SMAs) has also been included in this reprint.     Abstract: Toll-like receptors (TLRs) represent a powerful system for the recognition and elimination of pathogen-associated molecular patterns (PAMPs) from bacteria, viruses, and other pathogens and damage-associated molecular patterns (DAMPs) released from dying cells. Typical PAMPs include bacterial cell wall components, viral pathogens, or pathogenic nucleic acids, including viral RNA and DNA. Activation of TLRs leads to the production of proinflammatory cytokines and type I interferons which are important for the induction of the host immune response against bacterial and viral infections. However, dysregulation and overstimulation can be detrimental, leading to hyper-inflammation, sepsis, and loss of tissue integrity. The involvement of TLRs in inflammation and bacterial infection has been recognized for a long time. There is an increasing number of reports demonstrating the involvement of TLR activation in a variety of viral infections, associated with protective immunity, but also immune hyper activation and even viral replication. Recent data show the involvement of TLR activation in various acute respiratory viral infections, including SARS-CoV-2 and indicate an essential role in COVID-19 pathology. It aimed to gather newest data and hypotheses regarding molecular and cellular mechanisms of TLR triggering and activation and their downstream signaling pathways by viral infections, and their correlation to immunology and pathophysiology of the associated diseases, to facilitate translational research resulting in new targets for the treatment of viral infectious diseases including COVID-19

    Abstract: The papers collected in this Special Issue deal with the effects of ionizing radiation at molecular and cellular levels, as well as possible applications in medicine and radiation protection. In particular, some works analysed the early stages of the interaction between ionizing radiation and DNA focusing on DNA cluster damage, which is widely considered as an important step towards subsequent endpoints. Other studies investigated the consequences of hypoxia on the evolution of the initial DNA damage, and one work analysed genome-wide DNA alterations in irradiated human cells. Concerning radiation therapy, the response of cancer cells to different radiation types was characterized both in vitro and in vivo, and possible strategies to enhance such response were investigated; furthermore, FLASH radiotherapy was reviewed. In the framework of cancer therapy side effects, the consequences of high doses on gene expression in healthy cells were analyzed, and the RBE (Relative Biological Effectiveness) for late effects in the central nervous system was predicted by the BIANCA biophysical model. Finally, the protective role of melatonin and Vitamin D, as well as hypothermia, was reviewed, valine radiolysis was investigated, the changes in radio-sensitivity of embrios during the early developmental stages of the preimplantation were analyzed, and epigenetic modifications induced by ionizing radiation were reviewed

    Abstract: At the molecular level, the activation of growth factor receptors (GFRs) induces a mitogenic response and maintains cancer cell growth. The majority of malignant diseases are related to aberrant intra- and intercellular communication, associated with the GFR-mediated pathways. Moreover, the evasion of apoptotic signals and the requirement of angiogenesis were also found to be of fundamental importance for tumor progression and metastasis. In this context, a high expression of GFRs aids blood vessel formation, cell migration, and the inhibition of apoptosis. GFR-directed therapy that would theoretically selectively kill malignant cells and reduce the toxicity associated with nonselective conventional chemotherapy may be a promising treatment for cancer. Many intracellular proteins involved in GFR signal transduction can also function as oncogenes. Mutations affecting key proteins in the RAS/MAPK and PI3K/AKT pathways are known to be crucial in maintaining the malignancy of different types of cancers. This information has guided the development of compounds designed to target one or more of these pathways in cancer cells. Even though there have been important advances in our understanding of GFRs and their signaling, certain essential information is still lacking, and these membrane receptors are still being laboriously studied by several research groups, to find therapeutic solutions to unmet medical needs

In recent years, special attention has been focused on the possibility of giving materials new or improved properties by the introduction of nano or submicron size additives, fibers or whiskers. Using agents such as natural oils to enhance antimicrobial properties remains an exciting idea. Another area of research is the application of antibacterial monomers, which can be copolymerized in resins to kill oral pathogenic microflora. The use of new monomers or new compilations of various monomers to improve mechanical properties has also aroused interest. In addition, we are currently looking for new data regarding colonization of dental materials by pathogenic microbes and their influence on the other properties. Further, there are many new commercially available materials which should be investigated to verify their properties, which is important from the point of view of clinical practice. Original new research and reviews related to any of the topics mentioned above indicate the current directions of development of dental materials.     Abstract: Regarding the increasing use of nanomaterials in almost every area of our daily life, toxicological risk assessment is one of the major requirements for their safe handling. Especially at workplaces, inhalation is the major route of exposure and potential toxicity, and effects on the lung need to be considered. Furthermore, neurotoxicity associated with exposure to nanomaterials is a growing field of scientific investigation. However, not every single nanomaterial can be assessed in long-term animal inhalation studies, due to limited resources as well as political and societal efforts to reduce animal experiments according to the 3R principles. Thus, during the last few years, significant efforts have been dedicated to grouping nanomaterials, mainly based on advanced in vitro models. These new approach methodologies (NAMs) include detailed characterization of the respective materials in physiologically relevant media, but also more realistic exposure systems, such as co-cultures, also at the air–liquid interface, combined with comprehensive cellular investigations that provide quite detailed toxicological profiles. Thus, nanoparticles show differences in toxicity depending on their size, surface reactivity, and dissolution kinetics. Adverse cellular effects include inflammation, genotoxicity, oxidative stress, and epigenetic alterations. This Special Issue aims to highlight the recent advances in the mechanisms of nanomaterial toxicity, as well as the approaches for risk assessment, linking nanoparticle characteristics as well as in vitro toxicity to in vivo observations for advanced risk assessment

    Abstract: This Special Issue covers an important topic that has relevance not only from a scientific point of view but also from nutritional, policy, and industrial perspectives. We are currently being subjected to increasing obesogenic pressures, with sugars being hidden in many foods, creating the perfect environment for the slow but consistent progression of metabolic dysfunction. The consumption and impact of dietary sugars on the gastrointestinal system is discussed, together with the dietary sources of naturally occurring and added sugars, as well as the role of different diets and dietary regimens in preventing post-prandial sugar increases and hyperinsulinemia. The intestinal metabolism of dietary sugars, including the formation and absorption of advanced glycation end products and their impact on gut microbiota, is also considered. In terms of the impact of dietary sugars on other metabolic processes, the increased and sustained consumption of sugars causes long-term alterations in beta-cell function and the hypothalamic pathways controlling energy balance and the development of addictive behaviors, inducing mitochondrial impairment, oxidative stress, and the modulation of inflammatory pathways. Seeking to advance the development of prevention strategies to avoid the negative impact of dietary sugars, we outline the role of exercise in reducing blood markers of glucose dysmetabolism and the importance of preventing excessive sugar consumption in critical phases of development owing to its long-term consequences.
