Abstract: Musculoskeletal research deals with the effects of the orthopedic treatment of pathologies on the biomechanics of the affected areas and on the musculoskeletal system. Biomechanical measurement methods enable the quantitative determination of these influences and allow for an assessment of their extent and size for the patient (in vivo). The range of examination methods is particularly wide in this field of musculoskeletal research. On the one hand, in vitro examinations under laboratory conditions on simplified models, such as artificial bones or specimens from donors, will be implemented. With the help of these models, for example, new biomaterials or implants for the treatment of fractures are often examined for their primary stability or the influence of a joint replacement on the kinematics. In contrast to experimental in vitro studies, numerical methods will be increasingly applied to analyze a large number of implant configurations and loading scenarios. With the method of clinical motion analysis, a comprehensive view of the musculoskeletal system is performed directly in vivo on the patient. For example, it allows for the monitoring and control of therapeutic interventions. These are just a few examples from the field of musculoskeletal research and its methods. They all have the common goal of increasing patient safety. This Special Issue intends to provide the reader with an exciting overview of current research in the field of biomechanical investigations for the treatment of musculoskeletal diseases

Abstract: Herein we describe some molecular aspects of the recent anti-COVID-19 research, including some approaches making use of natural remedies, especially from medicinal plants, in silico methods, and different experimental procedures that led to the discovery of novel targets of SARS-CoV-2 and consequently novel potential COVID-19 therapeutics. COVID-19 vaccines also find mention in this book with also a description of some adverse effects reported in some cases. Considering that the different beta coronaviruses present certain conserved biomolecular mechanisms and, thus, some common therapeutic targets, finding effective therapies for a specific member of their family (such as SARS-CoV, SARS-CoV-2, and MERS-CoV) even in a scenario of a reduced impact at the time of the related disease on humankind is still relevant as it could offer or inspire efficacious therapeutic strategies not only in the context of novel pathogenic mutations of the already known beta coronaviruses, but also when future zoonoses caused by other beta coronaviruses will emergence, which is not unlikely to occur in the current world where the interactions between humans and wildlife are made more and more frequent due to the growing loss of intact ecosystems worldwide

Abstract: In order to build a promising future, health and sustainability must be interelated. Marine, forestry, agriculture, and food systems are important sources of bioproducts used in health applications. To explore the potential of such sources for the development of natural products capable of biological activities, it is necessary to develop new technologically sustainable strategies. Despite the range of natural compounds already available, there is a need to identify bioactive molecules (e.g., polysaccharides, proteins and peptides, polyunsaturated fatty acids, and polyphenols) from different natural sources with positive health properties, including antihypertensive, antidiabetic, anti-obesity, antimicrobial, anti-atherosclerotic, antioxidant, antithrombotic, immune-modulatory, relaxing, and satiety-inducing effects. The Second Edition of this Special Issue aimed to identify and gather works on the latest varied sources of bioproducts, the biological and functional activities of these bioactive compounds, their mechanisms of action, and the methods used for extraction and purification, without losing our focus on alignment with the concept of green technology

Abstract: Over the centuries, technological advancements have significantly enhanced various aspects of human life by improving the performance of materials that interact with living systems. Nevertheless, the mass production and disposal of numerous newly developed materials have given rise to a pressing 21st-century challenge: environmental sustainability. For instance, during the COVID-19 pandemic, the World Health Organization estimated that 89 million facial masks were required monthly for frontline health workers. A promising solution to this challenge lies in the micro- and nanofabrication of bio-based materials, which can transform current technology that is reliant on traditional materials by providing recyclable, biodegradable, and energy-efficient alternatives. Bio-based materials, such as papers and silk textiles, have demonstrated their sustainability over thousands of years. Recent progress in science and engineering has unveiled innovative functionalities and applications of these conventional materials through processes such as regeneration, recombination, and structural modification. This Special Issue is dedicated to exploring the future potential of bio-based materials in various applications interfacing with living systems, such as the human body, plants, soil microbiomes, and food

Abstract: Autophagy is a conserved degradative mechanism essential for cellular homeostasis in eukaryotic organisms. It is a cellular response mechanism that helps cells adapt and survive under various stressful conditions. It involves the degradation and recycling of cellular components to maintain cellular homeostasis and protect against damage from stressors like nutrient deprivation, oxidative stress, infection, and protein misfolding. However, the understanding of the molecular mechanisms of autophagy regulation is still in significant development, including the involvement of gasotransmitters and small signaling molecules, such as those produced under ROS, RNS and RSS. Dysregulation of autophagy has been implicated in various diseases, making it an important area of research to improve biomedical therapies and strategies for overcoming the challenges from environmental stresses. Therefore, this Special Issue aims to review the current developments regarding autophagy research

Abstract: Great strides have been made in the area of hydrogel science since the first hydrogels were described in the 1960s. Hydrogels usually consist of three-dimensional matrices of hydrophilic polymers, held together by chemical or physical crosslinks, or supramolecular assemblies of small amphiphilic molecules. The gelation process occurs in response to a physical or chemical stimulus, such as temperature, pH, electric or magnetic field, enzymatic modification, light, and others. Consisting of mainly water molecules, they represent a unique class of materials, with many applications such as cell therapeutics, cartilage/bone regeneration, sustained drug release and drug delivery systems, tissue engineering, and 3D bioprinting. Despite these great strides, there is still much more to discover in this area. This Special Issue is focused on the use of hydrogels in tissue and bone regeneration. Hydrogels are particularly suited for this purpose as their physical characteristics resemble that of the extracellular matrix; as such, they have found applications as an extracellular medium for cancer cells, stem cells, and neuronal cells. This Special Issue also includes research papers on the other biomedical applications of hydrogels

Abstract: This Special issue focuses of the most relevant advances in the field of inborn errors of immunity. Indeed, as immunology is rapidly progressing, with the constant discovery of new disease entities and the better characterization of molecular mechanisms and targeted therapies, a continuous update is mandatory for clinician and researchers. In this Issue, the non-infectious manifestations of inborn errors of immunity (such as autoimmunity, atopy, and lymphoproliferation) are deeply discussed, as well as the impact of the novel genetic sequencing techniques in the diagnostic approach. Moreover, some well-known disorders, including common variable immunodeficiency, are extensively reviewed and revisited in a modern way. Also, diseases of recent description, including some disorders of immune dysregulation and inborn errors of immunity associated with syndromes, are described. Finally, some of the papers of this Special Issue focus in a detailed way the molecular mechanisms leading to immunodeficiency and immune dysregulation, also opening the way for further clinical and preclinical studies. As the expanding availability of immunological and genetic testing offers the opportunity to identify new disease entities and elucidate the function of new genes involved in the development and regulation of the immune response, we aim to present the most relevant innovations in this delicate field

Abstract: Around 20%–30% of nonmetastatic prostate cancer patients have high-risk localized disease. The best treatment for high-risk localized disease is still unclear, and the reliability of the definition of “high-risk” may need to be validated first. Although recent advances in radiotherapy for prostate cancer have yielded excellent long-term results, even in high-risk localized disease, there are no studies directly comparing it with local treatment options, such as prostatectomy. To address current challenges in high-risk localized and locally advanced prostate cancer, 12 original and 3 review articles covering aspects from basic research to clinical trials are published in this Special Issue

Abstract: It was first suggested nearly 50 years ago by Meyer-Rochow in Australia that the use of insects as food and feed sources could ease the problem of global food shortages, and that the WHO and the FAO should support the promotion of insects as a food item, especially in countries with a long tradition of entomophagy. However, food shortages in some parts of the world are only one aspect to consider; overeating and obesity in other parts of the world are another aspect. An increased use of edible insects might well be able to assist in overcoming, at least to some extent, both of these problems. This Special Issue of Foods represents Volume 2 of the topic “Edible Insects as Innovative Foods: Nutritional, Functional and Acceptability Assessments”. Some of the 20 contributions by scientists from 13 different countries deal with hitherto unreported food insects, and others explore the effects that a diet containing insects or insect products can have on the gut microbiota of the consumer, whether human or non-human. Food safety questions are not ignored, and questions related to the chemical composition of food insects, their content in terms of nutrients and antinutrients, and their acceptability by consumers are additional topics that the articles in this book explore. Altogether, this reprint provides convincing reasons for the advantages to health and the environment that a greater use of insects as food and feed would present, and ends with the slogans “Forget about the pork and put a cricket on your fork!” and “Mealworms and spaghetti is food that makes you happy!”

Abstract: This second volume on the subject of intervertebral disc (IVD) regeneration ranges from in vitro IVD cell culture studies on cytokines, such as TGFβ3, GDF5, GDF6, ANG1, and ANG2, to diffuse idiopathic skeletal hyperostosis (DISH) to novel advances in the diagnosis of painful discs in reports using natural language processing (NLP). This diverse issue consists of seven articles that provide a comprehensive overview of recently applied research insights. Three original articles focus on the effects of cytokines in primary cell culture. Then, two original articles report on transcriptomics data, i.e., single-cell RNA-sequencing data of in vivo rat animal models of IVD degeneration, and human prime PCR data comparing trauma versus DISH-IVDs. One original article addresses the correct diagnosis of lumbosacral transitional vertebrae (LSTV) in scoliotic patients, and another original article addresses the usage of NLP to improve the diagnosis of painful IVDs. The Special Issue is concluded with a review on the importance of the relatively recent “hot topic” of extracellular vesicles (EVs) for regenerative purposes in IVDs

Abstract: Interactions between nucleic acids and proteins are essential requirements for the viability of cellular life because they are indispensable for many basic biological processes. DNA is typically presented as a specific double-stranded helical structure, but nucleic acids have great structural flexibility. Current knowledge demonstrates that the structural conformations of nucleic acids play critical roles in protein–DNA interactions. This book presents a collection of research findings published in the Special Issue of the International Journal of Molecular Sciences, titled “Impacts of Molecular Structure on Nucleic Acid-Protein Interactions”.The breadth of research findings reported here demonstrates that the structural flexibility of nucleic acids plays critical roles in their interactions with proteins, with important implications across a range of human diseases, including cancer and some infectious diseases.Prof. Richard BowaterUniversity of East Anglia, Norwich, United KingdomProf. Václav BrázdaInstitute of Biophysics of the Czech Academy of Sciences, Brno, Czech Republic

Abstract: Biocatalysis is a sustainable alternative for the chemical industry in manufacturing, monitoring, and waste management. Biocatalytic processes perform with isolated enzymes or whole cells as biocatalysts. Whole-cell biocatalysts offer some unique advantages of cascade reactions catalyzed by multienzymes as well as a single bioredox reaction with cofactor regeneration in a single strain. Therefore, whole-cell biocatalysts are widely applied for biosynthesis/biotransformation to produce value-added chemicals as well as the complete mineralization of organic pollutants.Biological catalytic processing using whole-cell biocatalysts includes biocatalyst engineering, bio-reaction engineering, and downstream processing. In addition to the traditional screening of microbial strains and immobilized whole-cell biocatalysts, modern genetic engineering, metabolic engineering, and synthetic biology make tailored whole-cell biocatalysts possible. At the same time, some integrated processes have successfully been applied in the catalytic processing using living whole-cell biocatalysts, such as harnessing biocompatible chemistry to interface with the microbial metabolism as well as using various separation techniques for in situ product removal. This reprint on “Microbial Biocatalysis” provides a comprehensive overview of the recent developments of catalyst discovery, catalyst modification, and process intensification for whole cell catalysis in fermentation, biotransformation or biodegradation processes

Abstract: Psychiatric disorders in children and adolescents are relevant both in terms of their prevalence (they are roughly estimated to affect about 20% of school-aged children and adolescents) and of their impact on the quality of life of the child/adolescent and their family, but also on the society as a whole. Disorders affecting children and adolescents are increasingly seen as having highly specific features, and, therefore, require an equally specific approach despite the coexisting need to integrate contributions from other professions (e.g., pediatricians, psychiatrists, psychologists, and therapists of different sorts). However, the available data are still insufficient, especially for treatment options and too often therapeutic choices are still made based on studies conducted in adults. Even more important, the existence itself of some disorders is questioned especially outside the scientific field (e.g., ADHD). This Special Issue provided cutting edge data on different aspects of child and adolescent psychiatry, including etiopatogenesis, clinical characteristics and diagnosis, medical and neurological comorbidities (and psychiatric comorbidities of medical and neurological disorders), impact on patients/families/society (including school and other social groups), prognosis, and treatment options of the different disorders

Abstract: In recent decades, independent national and international research programs have revealed possible reasons behind the death of managed honey bee colonies worldwide. Such losses are not due to a single factor, but instead are due to highly complex interactions between various internal and external influences, including pests, pathogens, honey bee stock diversity, and environmental changes. Reduced honey bee vitality and nutrition, exposure to agrochemicals, and the quality of colony management contribute to reduced colony survival in beekeeping operations. Our Special Issue (SI) on ''Monitoring of Honey Bee Colony Losses" aims to address the specific challenges that honey bee researchers and beekeepers face. This SI includes four reviews, with one being a meta-analysis that identifies gaps in the current and future directions for research into honey bee colonies' mortalities. Other review articles include studies regarding the impact of numerous factors on honey bee mortality, including external abiotic factors (e.g., winter conditions and colony management) as well as biotic factors such as attacks by Vespa velutina and Varroa destructor

Abstract: Ornamental crops account for more than US $42 billion globally. With the exception of few floral species, limited genetic, genomic, and breeding information is publicly available, owing to the fact that the majority of breeding work is performed by the private sector. Public research programs are increasingly participating in ornamental cultivar development and genetic studies. With lower sequencing costs, genomic information of non-model species including ornamental crops is continuously becoming available. Ornamental breeding utilizes a wide array of breeding strategies ranging from traditional crossing and selection methods to the use of next-generation sequencing in genomics and transcriptomics for gene identification and trait development. A continuing search of new species for the ornamentals industry has resulted in the utilization of tools that increase diversity and in the development of alternative methods for obtaining new crops by achieving interspecific and intergeneric crosses. This Special Issue aimed to present papers on new breeding methods, novel cultivars and species entering the ornamental industry, the identification of genes conferring novel traits, technological developments in ornamentals research, and the use of next-generation sequencing to improve ornamental plants

Abstract: Dear colleagues, This Special Issue, "Endothelial Dysfunction: From Pathophysiology to Novel Therapeutic Approaches", focuses on the pathophysiology of endothelial dysfunction, new biomarkers for endothelial dysfunction related to cardiovascular disorders or tumors, and novel therapeutic approaches for endothelial dysfunctions. Vascular endothelium is an active tissue and plays a crucial role in the maintenance of vascular homeostasis. Chronic exposure to risk factors, such as hypertension, high cholesterolemia, or oxidative stress, induces endothelial dysfunctions and results in a loss of endothelial integrity, smooth muscle cell proliferation, and macrophage recruitment. The pathophysiology of endothelial dysfunction (ED) is complex and multi-factorial factors are involved, such as oxidative stress or chronic inflammation. The primary prevention of cardiovascular risk factors and endothelial dysfunctions, as well as the early detection of or molecular imaging techniques for endothelial dysfunction, helps to prevent the development of cardiovascular disorders. Novel therapeutic approaches or drug delivery systems for endothelial dysfunctions have had promising beneficial effects in preclinical or clinical levels by affecting the progression of atherosclerotic changes, tumor angiogenesis, and host-immune reactions near tumor environments

Abstract: Food waste is becoming an important and growing concern at both local and global levels. According to the Food and Agriculture Organization of the United Nations (FAO), one-third of all food production is wasted globally, and in particular, 1.3 billion tons of food produced for human consumption is wasted per year, representing an economic loss of EUR 800 billion. The main foods wasted are represented by vegetables, fruits, meat, and fish. Considering the high availability and the composition of food waste, there is an increasing interest in their bio-valorization. Moreover, according to the global Sustainable Development Goals (SDGs 12 and 13), an appropriate waste management represents an essential prerequisite for the sustainable development. This reprint collects interesting manuscripts regarding innovative research focused on food waste valorization through fermentation processes for obtaining value-added products such as enzymes, feed additives, biofuels, animal feeds as well as other useful chemicals or products, food-grade pigments, and single-cell protein (SCP), enhancing food security and environmentally sustainable development
