    Abstract: Penthorum chinense Pursh (Penthoraceae) has been used as a Miao ethnomedicine for the treatment of jaundice, cholecystitis, edema, infectious hepatitis and anti-drunk hangover in China. The aim of present study is to investigate the possible protective effects of Penthorum chinense against chronic ethanol-induced liver injury. Mice were fed a Lieber-DeCarli liquid diet containing alcohol or isocaloric maltose dextrin as control diet with or without aqueous extract of Penthorum chinense (PCP, 5.15 and 10.30 g/kg/BW) for 4 weeks. Silymarin (86 mg/kg) was used as positive control to compare the efficacy of PCP against chronic ethanol-induced hepatotoxicity. Treatment with PCP (10.30 g/kg) significantly reduced the increases in serum ALT and AST levels, hepatic lipid accumulation and inflammatory cytokines (i.e. TNF-α, IL-6), which were induced by chronic ethanol exposure. PCP was also found to attenuate reactive oxygen species (ROS) generation and malondialdehyde (MDA) level, restore the glutathione (GSH) depletion, and increase the superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities. In addition, PCP supplementation (10.30 g/kg) inhibited the induction of hepatic cytochrome P450 2E1 (CYP2E1), a major contributor to ethanol-mediated oxidative stress, and up-regulated the expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and its downstream anti-oxidant protein heme oxygenase-1 (HO-1) in ethanol-treated mice. These results indicate that the co-treatment with aqueous extract of Penthorum chinense (10.30 g/kg) protects against chronic ethanol-induced liver injury, possibly through suppressing CYP2E1-mediated oxidative stress and enhancing the oxidant defense systems via the activation of Nrf2/HO-1 pathway.

    Abstract: Glycyrrhizae radix (GR) is a medicinal herb extensively used in traditional Chinese medicine. This study aimed to evaluate the pharmacological effect of GR and the possible mechanisms of GR, to provide a pharmacological basis in traditional medicine. In the present study, C. elegans (L1-larvae to young adults) was exposed to 0.12-0.24g/mL of GR in 12-well sterile tissue culture plates at 20°C in the presence of food. Lethality, growth, lifespan, reproduction, locomotion, metabolism, intestinal autofluorescence, and reactive oxygen species (ROS) production assays were performed to investigate the possible safety profile and beneficial effects of GR in these nematodes. We found that the lifespan of nematodes exposed to 0.18-0.24g/mL of GR was extended. We then determined the mechanism of the longevity effect of GR using quantitative reverse transcription PCR and oxidative stress resistance assays induced by heat and paraquat. Prolonged exposure to 0.12-0.24g/mL of GR did not induce lethality, alter body length, morphology or metabolism, affect brood size, locomotion, the development of D-type GABAergic motor neurons, or induce significant induction of intestinal autofluorescence and intestinal ROS production. In C. elegans, pretreatment with GR suppressed the damage due to heat-stress or oxidative stress induced by paraquat, a ROS generator, on lifespan, and inhibited the induction of intestinal ROS production induced by paraquat. Moreover, prolonged exposure to GR extended lifespan, increased locomotion and decreased intestinal ROS production in adult day-12 nematodes. Furthermore, prolonged exposure to GR significantly altered the expression patterns of genes encoding the insulin-like signaling pathway which had a key role in longevity control. Mutation of daf-16 gene encoding the FOXO transcription factor significantly decreased lifespan, suppressed locomotion, and increased intestinal ROS production in GR exposed adult nematodes. GR is relatively safe and has protective effects against the damage caused by both heat-stress and oxidative stress at the examined concentrations. Furthermore, GR is capable of extending the lifespan of nematodes, and the insulin-like signaling pathway may play a crucial role in regulating the lifespan-extending effects of GR.

    Abstract: The cold/hot property of Chinese materia medica (CMM) and the application of its corresponding knowledge in the diagnosis, differentiation and treatment of diseases have been considered to be the extremely important part of traditional Chinese medicine (TCM). As highly abstracted TCM theory, the cold/hot property of CMMs is still not fully understood and remains to be elucidated by systems biology approach. The cold and hot properties of CMM are mainly defined by the response of the body to a given CMM. Metabolomics is a promising systems biology method to profile entire endogenous metabolites and monitor their fluctuations related to an exogenous stimulus. Thus, a metabolomics approach was applied to characterize the cold and hot properties of CMMs. Mice were intragastrically administered three selected cold property CMMs (i.e., Rheum palmatum L., radix et rhizoma; Coptis chinensis Franch, rhizome and Scutellaria baicalensis Georgi, radix) and three hot property CMMs (i.e., Cinnamomum cassia (L.) J. Presl, cortex; Zingiber officinale Roscoe, rhizoma and Evodia rutaecarpa (Juss.) Benth., fructus) once daily for one week. The comprehensive metabolome changes in the plasma of mice after treatment with cold or hot property CMMs were characterized by ultra-high performance liquid chromatography/time of flight mass spectrometry (UHPLC/Q-TOF-MS), and the potential biomarkers related to cold and hot properties of CMM were explored. Metabolites perturbation in plasma occurs after treatment with cold CMMs and hot CMMs in mice, and 15 and 16 differential biomarkers were identified to be associated with the cold and hot properties of CMMs, respectively. Among them, LPC (18:0), LPC (18:1), LPC (20:4) and LPC (20:5) showed decreased trends in the cold property CMM treated groups, but increased in the hot property CMM treated groups. There is a strong connection between the cold/hot property of CMMs and lysophosphatidylcholines metabolism. This study offers new insight into CMM properties and their clinical application.
